It depends...if you've got a custom rom on there now (meaning you've run the titan unlocker before) then you will have to re-lock it. Otherwise, you should be ok.

I have custom ROM from DCD's kitchen over at XDA. So I re-lock it then run as a normal update for the desktop without using the bootloader?

Yep! Relock it using the relocker SPL (pay close attention to EVERY instruction) and then once finished, you will be in the relocked bootloader. Once there, you can start the RUU from the zip file. Don't worry if you get stuck, it's really hard to mess it up. Just be patient. The installer gave me some issues, but with patience, you'll be just fine.
