I know that for me it depends which USB port I plug into since my work and home pc have multiple ports.
Try changing to a differnet port. Try deleting and reinstalling ActiveSync. Also, Sprite allows you to backup your device to your storage card...so try that option. Can you open ActiveSync on your pc? Do you have a firewall that's keeping you out? Give us a bit more info. late, Coz

Something that just happened to me with the same problem, was that I updated one care and it reset my firewall settings. It took me about an hour to figure it out. I don't know if that helps, but if you updated it could be culprit.
