Since none of the filenames are changed and no files were added or removed, I don't think you need to rebuild the DSM, but it won't hurt anything.

It may be better to copy one of the existing folder sets a then modify your copy. There are two folders that make up a theme...
Optional_XXX_Base_DPI_96
Optional_XXX_Redist_DPI_96
In this case you'll need to regen the GUIDs, rebuild the DSMs in both folders, and update the option.xml in the "Optional_XXX_Base_DPI_96" folder.

Unfortunately I'm not sure about the sound. Hopefully someone else can chime in.

If you are wanting to change the sound the phone makes on bootup, you can replace the "phoneOn.wav" file in the sprint customizations folder with one you want. Just have to name the file the same. Optionally you can do the same with the phoneOff.wav file.
