That really depends on what RUU (ROM Upgrade Utility) you use to flash the ROM. If it's the normal one then there's no need to put in bootloader first. If it's the NOID one then yes you should put the phone in BL.
But if it's easier, you can just always put the phone in bootloader mode--that should work with any of the RUU apps.
