This may be an obvious response, but might be worth trying. I've had a similar issue before, and re-booting the computer has helped one time. The other time it happened, i ended up having to switch to a different USB port.
For some reason, that cleared everything up, and then i could switch back to the original USB port and everything worked.
The other option might be to delete the relationship in Activesync or Mobile Connect, restart phone and computer, and see if that helps.
