My final words on the subject:
1) Full capcity of Li-Ion is available immediately. In fact, available capacity decreases due to increasing internal resistance per charge cycle.

2) I wouldn't trust a manual for an aftermarket battery... and I haven't read any manuals for Li-ION batteries that suggest discharge cycles. The most I've seen them suggest is that the gauge may not be calibrated until a full discharge cycle, because the battery needs to measure timing accoring to your use... Note that this is only useful for devices that have a time remaining gauge... which phones don't have.

3) Li-ion batteries come charged for a reason, because if they are stored with no charge, they will quickly be damaged.

4) The biggest enemies of Li-Ion battery life are time, temperature, and discharge cycles, and depletion. This means - don't leave your battery lying around at a high temperature, use your charger when you can (but don't charge your phone for small periods of time, this raises the temperature, only charge when you know for SURE that you have time to fully charge the battery), and don't do wacko things like "deep discharge" your batery, because you WILL irreversibly damage the Li-Ion cells. Especially dont let the voltage drop below the threshold. Also, don't store the battery at 100% charge, because this will reduce the life as well.

I want to re-iterate that the reason you leave it on the charger when you can is that discharge seems to produce a significant amount of heat due to internal resistance, which probably screws with the battery life, Even though you will be storing the battery at 100% as a result, this is better than generating heat and discharging the battery. Of course, if you are going to store this as your spare battery, discharge it to 40%, then store in a cool place!
