Yes you should do a nandroid after rooting your phone!

A nandroid is a back-up of your phones software at the time that you do the nandroid, so if you have a custom rom on your phone and do a nandroid then that's what gets backed up, it's just like a restore point on a computer.

Root your phone and then do a nandroid and then you will have that back-up in case you need to return the phone to stock <or to the last rom you was using in case you want to return to the last rom you was running> and take it in for service, really the most important part of the nandroid back-up is the backing up of your wimax keys <4g keys> because those can't be replaced, , and that's done when you nandroid the phone. It's not part of the rooting process so like I said after you have ran Revolutionary and rooted your phone boot the phone into recovery and do a nandroid and then feel free to start flashing custom roms.
